科學家：茲卡病毒確實造成腦細胞癱瘓、死亡 與小頭症關聯尚待釐清

透過蚊子傳染的茲卡病毒蔓延拉丁美洲，巴西2015年5月開始爆發疫情，超過5000個嬰兒出生時出現小頭症與嚴重發育遲緩的症狀，但卻無法證實茲卡與小頭症的關聯。4日科學家首次提出生物性直接證據，根據實驗觀察發現，茲卡病毒會鎖定傷害生長中的腦細胞，進而破壞或癱瘓其運作。

「這是第一個直接證據！」此項研究也在幹細胞研究權威期刊《Cell Stem Cell》發表，約翰霍普金斯大學細胞工程學院（Johns Hopkins'Institute for Cell Engineering）神經學教授明國莉表示，在此次發現以前皆為間接證據，沒有直接的實驗佐證。研究提供了兩者之間可能的病理解釋，有助於發展針對茲卡病毒感染的治療方法。但此僅為證實茲卡病毒與小頭症的第一步，尚未完全釐清兩者的絕對關聯。

「茲卡病毒會先殺死大腦中的神經祖細胞。」科學家在實驗室中將大腦最重要的兩種細胞－神經幹細胞與神經祖細胞感染茲卡病毒，而發現在3天之內，細胞的85%被其破壞感染。若病毒無法殺死神經細胞，也將使其運作趨緩。





神經祖細胞類似幹細胞，但他卻更加特殊，由於幹細胞能透過無限再生來保持運作，但祖細胞卻有再生次數的限制，這代表若其中一個祖細胞被病毒破壞，將無法透過再生來修補缺陷。

茲卡病毒有特別針對神經祖細胞攻擊的能力，侵入細胞並使用細胞再生的功能，複製病毒傳遞到健康的細胞中。且受感染的細胞分裂速度較一般的細胞慢，這也可能是造成畸形的原因。





雖80%的茲卡病毒感染者都不會發病，但若孕婦感染茲卡病毒卻會造成胎兒的變異。在2015年9月的一項研究中，觀察了88位孕婦，其中有72名感染了茲卡病毒，而另外16名則呈現陰性反應。感染的婦女中有3分之1的胎兒呈現異常狀態。「這是非常高的比例。」共同研究者加州大學洛杉磯分校小兒傳染專家尼爾森（Karin Nielsen）表示，不能再忽視這樣的問題。
